Naturally Occurring p16Ink4a-Positive Cells Shorten Healthy Lifespan

Introduction

The investigators employed a novel transgenic INK-ATTAC murine model to elucidate the physiological aspects and consequences of naturally occurring senescent cells. This research paper describes the anti-aging effect AP20187-based p16-positive cells clearance and the perspective of employing the treatment to annihilate age-related diseases.

Key findings

  • p16Ink4a-positive cells were determined as generators of adipose tissue formation disruption, cataract formation, cardiac stress tolerance deterioration and glomerulosclerosis.
  • INK-ATTAC related AP20187 injection was used to promote apoptosis of p16Ink4a-positive cells of wild type mice from two cohorts with different diet, genetical background and dosing schedule.
  • Clearance of p16Ink4a- expressing cells had an anti-tumorigenic potential and diminished senescence-associated detrimental effect on observed organs (kidney, heart, fat and eye) without any vital functions disruption.
  • AP-treatment was ineffective when administered to unmanipulated mice.
  • AP-treatment seemed to deteriorate wound healing process.

Conclusion

p16Ink4a-expressing cells accumulation has detrimental effects on proper organs function in murine models. AP-based treatment in ATTAC mice has a great potential in diminishing age-related changes. However, it is still not clear, how AP influences wound healing and what is the most effective endurance of AP injections administration to attain the best results.
